The effect of Iron and H2O2 on plasma membrane of wheat roots studied by Fourier Transform Infrared Spectroscopy |

摘 要:We studied the alterations in structure and function within the plant plasma membrane (PM) caused by treatment with H2O2, FeSO4, and FeCl3 using attenuated total reflectance infrared (ATR-IR) and fluorescence spectroscopic techniques. It was observed that the 200 μM H2O2, FeSO4, and FeCl3 treated PM showed a decline in H+-ATPase activity and reduced membrane fluidity. Computer-aided analyses the FTIR spectrum indicated that the content of the α-helices decreased and βsheet increased in the secondary structures of proteins after exposure to the oxidants 200 μM H2O2, FeSo4, and FeCl3. The number of P=O and C=C bonds in the lipids of the membrane rapidly declined. These were the main reasons for the decline in H+-ATPase activity and the reduced fluidity of the PM. |
